Mayers became the 14th West Indies player to score a century on debut as Bangladesh failed to pick up a wicket in the second session. West Indies were left needing 129 in the final session.
Bonner made 86 but fell in the first over of the final session. His dismissal also brought an end to the 216-run fourth-wicket stand.
Mayers struck unbeaten 210 to carry West Indies to a sensational come-from-behind three-wicket win.
Mayers and Bonner helped West Indies pull off the fifth-highest chase in Test cricket and also the highest in Asia.
